Biological functions and therapeutic potential of acylation by histone acetyltransferases

نویسندگان

چکیده

Histone lysine acylation is a major class of histone post-translational modifications involved in essential biological activities, such as transcriptional regulation, DNA-damage repair, and cell-cycle progression. Abnormal strongly associated with various diseases, cancer. Thus, acetyltransferases (HATs), the “writers” that catalyze acylation, are promising targets for cancer treatment. Rapid developments structural biology artificial intelligence have facilitated development drugs targeting HATs. To provide new ideas exploring novel HAT modifiers high efficiency selectivity, this article reviews relationships between illustrates catalytic mechanisms through biology, summarizes research progress modifiers.

Microarray Analysis Reveals Potential Biological Functions of Histone H2B Monoubiquitination

Histone H2B monoubiquitination is a key histone modification that has significant effects on chromatin higher-order structure and gene transcription. Multiple biological processes have been suggested to be tightly related to the dynamics of H2B monoubiquitination. However, a comprehensive understanding of biological roles of H2B monoubiquitination is still poorly understood. In the present stud...

Regulating histone acetyltransferases and deacetylases.

Histone acetyltransferases and histone deacetylases regulate the acetylation of histones and transcription factors, and in doing so have major roles in the control of cell fate. Many recent results have indicated that their function is strictly regulated in cells through the modulation of their levels, activity and availability for interaction with specific transcription factors. In this review...

Histone Acetyltransferases in Plant Development and Plasticity

In eukaryotes, transcriptional regulation is determined by dynamic and reversible chromatin modifications, such as acetylation, methylation, phosphorylation, ubiquitination, glycosylation, that are essential for the processes of DNA replication, DNA-repair, recombination and gene transcription. The reversible and rapid changes in histone acetylation induce genome-wide and specific alterations i...

Chromatin: Histone acetyltransferases in control

Research on transcriptional control in eukaryotes traditionally takes a two-pronged approach, relying on genetic analysis to define the regulatory elements and transcription factors required to activate or repress a promoter, and on biochemical methods to purify the transcription factors, define their structure and reconstruct their activities in vitro. Genetics usually leads the way, but occas...

Prediction of Biological Functions by Histone Modification Patterns Profiling

Histone modifications provide an important layer of gene regulation in eukaryotes. In this paper, we propose an approach that identifies the histone modification patterns most relevant for specific biological functions, such as flowering in plants. We first propose a new pattern scoring method, which evaluates the importance of each combinatorial pattern of histone modifications; this is used a...
